In situ microscopic investigation of plant cell walls deconstruction in biorefinery

چکیده
Plant biomass that primarily composed of polysaccharides and lignin constitutes a renewable energy source. The conversion of biopolymers into fuels, energy, and chemicals with a concept of biorefinery involves a chemical/biological treatment to facilitate components release performance. However, the accurate understanding of cellular architecture of plant cell walls remains unclear owing in part to complexity of biomass chemical structures as well as limitations of traditional analytical methods that may bring irreversible modifications. In situ microscopic techniques are readily required to visualize, characterize, and quantify the dynamic features of plant cell walls following treatments on multi-scale levels. This review gives insights into the recent contributions of a series of non-destructive techniques including confocal laser scanning microscopy, confocal Raman microscopy, FT-IR imaging microscopy, and atomic force microscopy to the field of biorefinery, focusing on the deconstruction of plant tissues. We also highlight the critical outlook and scientific challenges on the study of biomass utilization as well as microscopy applications.

In situ label-free imaging of hemicellulose in plant cell walls using stimulated Raman scattering microscopy
BACKGROUND Plant hemicellulose (largely xylan) is an excellent feedstock for renewable energy production and second only to cellulose in abundance. Beyond a source of fermentable sugars, xylan constitutes a critical polymer in the plant cell wall, where its precise role in wall assembly, maturation, and deconstruction remains primarily hypothetical. Effective detection of xylan, particularly by...

متن کاملDirected plant cell-wall accumulation of iron: embedding co-catalyst for efficient biomass conversion
BACKGROUND Plant lignocellulosic biomass is an abundant, renewable feedstock for the production of biobased fuels and chemicals. Previously, we showed that iron can act as a co-catalyst to improve the deconstruction of lignocellulosic biomass. However, directly adding iron catalysts into biomass prior to pretreatment is diffusion limited, and increases the cost of biorefinery operations. Recent...
متن کاملVisualization of Miscanthus × giganteus cell wall deconstruction subjected to dilute acid pretreatment for enhanced enzymatic digestibility
BACKGROUND The natural recalcitrance of lignocellulosic plant cell walls resulting from complex arrangement and distribution of heterogeneous components impedes deconstruction of such cell walls. Dilute acid pretreatment (DAP) is an attractive method to overcome the recalcitrant barriers for rendering enzymatic conversion of polysaccharides. In this study, the internodes of Miscanthus × gigante...
